目的:探讨葛根素对雷帕霉素造成人脐静脉内皮细胞(HUVEC)损伤的影响及其可能机制。方法:培养HUVEC并分组:对照组,葛根素组,雷帕霉素组,葛根素干预组。各组培养48h后,检测各组HUVEC的增殖,NO浓度及凋亡率的情况。结果:与对照组比较,葛根素组NO浓度明显升高(P<0.05),HUVEC凋亡率明显降低(P<0.01);葛根素干预组NO浓度明显降低(P<0.01);雷帕霉素组可抑制HUVEC增殖,NO浓度明显降低,HUVEC凋亡率明显升高(P<0.01)。与雷帕霉素比较,葛根素组NO浓度明显升高,HUVEC凋亡率明显降低(P<0.01);葛根素干预组促进HUVEC增殖,NO浓度明显升高,HUVEC凋亡率明显降低(P<0.05,P<0.01)。结论:葛根素对雷帕霉素造成HUVEC损伤具有一定的拮抗作用。
Objective: To investigate the effect of puerarin on human umbilical vein endothelial cells (HUVEC) induced by rapamycin and its possible mechanism. Methods: HUVECs were cultured and divided into control group, puerarin group, rapamycin group and puerarin intervention group. After 48 hours of culture, the proliferation, NO concentration and apoptosis rate of HUVEC in each group were detected. Results: Compared with the control group, the concentration of NO in puerarin group was significantly increased (P <0.05), the apoptosis rate of HUVEC was significantly decreased (P <0.01), while the concentration of NO in puerarin group was significantly lower (P <0.01) Supramolecular group could inhibit the proliferation of HUVEC, the concentration of NO was significantly decreased, and the apoptosis rate of HUVEC was significantly increased (P <0.01). Compared with rapamycin, the concentration of NO in Puerarin group was significantly increased, and the apoptosis rate of HUVEC was significantly decreased (P <0.01). The Puerarin intervention group promoted the proliferation of HUVEC, the NO concentration was significantly increased, and the apoptosis rate of HUVEC was significantly decreased <0.05, P <0.01). Conclusion: Puerarin can antagonize the injury of HUVEC induced by rapamycin.
