Berenblun 及Shubik 最早提出的致癌作用的两期概念在实验肿瘤研究的基础上已逐渐得到广泛的承认。他们的研究是在鼠皮肤上进行的,结果提示一个特殊的不可逆的激发期,此时正常细胞变成潜在性肿瘤细胞;以后是促进期,在促进期内,这种细胞被刺激发展成真正的肿瘤。激发期要有一种化学性致癌物如9,10—二甲—1,2苯并蒽(DMBA),而促进剂可以是无特殊刺激的如巴豆油,它不是一种致癌剂。Ritchie 及Salffiotti 证明皮肤致癌剂两个时期,用2-乙酰氨基芴为激发剂,巴豆油为促进剂。他们也证明在用DMBA 作激发剂时,2-乙酰氨基芴可作为促进剂。Solt 及Farber 在制成的模型中研究
The two concepts of carcinogenesis first proposed by Berenblun and Shubik have gradually been widely recognized on the basis of experimental tumor research. Their study was performed on mouse skin and the results suggest a special irreversible provocative phase where normal cells become potential tumor cells; later, during the promotion phase, the cells are stimulated to develop into true Tumor. There should be a chemical carcinogen such as 9,10-dimethyl-1,2benzopyrene (DMBA) during the challenge period, and the accelerator may be croton oil without special stimuli. It is not a carcinogen. Ritchie and Salffiotti proved skin carcinogens two times, using 2-acetylaminostilbene as an activator and croton oil as an accelerator.
