目的研究和比较两种鹿鞭水提物对顺铂(CDDP)诱导小鼠急性肾损伤保护作用,并探讨可能机制。方法 64只雄性小鼠随机均等分为空白对照组、CDDP模型组、新西兰马鹿鞭和梅花鹿鞭水提物高、中、低剂量(100μg/g、200μg/g、400μg/g)+CDDP保护组;各鹿鞭保护组连续灌胃7天,在第5天给药1 h后(除空白对照组)小鼠均腹腔注射CDDP(20μg/g)诱导急性肾损伤。造模48 h后,测定小鼠血清和肾组织中肾功能指标水平和观察HE染色肾脏组织病理学切片变化。结果与CDDP模型组比较,两种鹿鞭水提物各剂量保护组均可改善肾功能指标水平和肾损伤状况,梅花鹿鞭较新西兰马鹿鞭效果更佳,其中梅花鹿鞭水提物高剂量保护组效果最为显著:BUN、CRE和MDA显著降低(P<0.01),SOD和GSH显著升高(P<0.01);明显改善损伤肾脏中肾小管变性和坏死的现象,但未恢复正常水平。结论两种鹿鞭水提物均对顺铂诱导小鼠急性肾损伤有明显保护作用,可能与清除自由基和抗氧化应激作用有密切联系。
Objective To study and compare the protective effects of aqueous extract of Cervus nipponense on acute kidney injury induced by cisplatin (CDDP) in mice and to explore the possible mechanism. Methods Sixty-four male mice were equally divided into blank control group, CDDP model group, high, medium and low dose (100μg / g, 200μg / g, 400μg / g) CDDP protection The rats in each group were given gavage for 7 days continuously. After 1 day of administration on the fifth day (except the blank control group), all mice were injected intraperitoneally with CDDP (20μg / g) to induce acute renal injury. After modeling for 48 h, the levels of renal function in serum and kidney of mice were determined and histopathological changes of kidney were observed. Results Compared with the CDDP model group, each dose protection group of two kinds of Lübian water extract could improve the level of renal function and renal injury, the Sliver whip better than New Zealand horse deer whip, which Sika deer whip high dose protection The results were as follows: BUN, CRE and MDA significantly decreased (P <0.01) and SOD and GSH significantly increased (P <0.01); renal tubular degeneration and necrosis were significantly improved, but did not return to normal level. CONCLUSION Both aqueous extract of Cervus nipponense could significantly protect against acute renal injury induced by cisplatin in mice, which may be closely related to the elimination of free radicals and antioxidative stress.
