筛选不同表型的胃癌细胞亚株。方法:用体外粘附法筛选对层粘素(laminin,Lin)有不同粘附力的MKN-45胃癌细胞亚株,并用Boyden小室法和裸鼠接种法观察细胞亚株的体内、外侵袭转移能力。结果:经体外反复以层粘素粘附筛选,得到高粘附力的MKN-45Lm+细胞亚株和低粘附力的MKN-45Lm-细胞亚株。Lm+细胞体外对人工基膜Matrigel的侵袭、移行能力及在裸鼠体内的成瘤性和肺转移灶形成率均大于Lm-细胞,并有高转移和高侵袭倾向。结论:体外粘附法能成功筛选特定表型的胃癌细胞亚株,胃癌细胞的粘附力与侵袭转移相关。
Screen different subtypes of gastric cancer cells with different phenotypes. Methods: MKN-45 gastric cancer cell sub-strains with different adhesion to laminin (Lin) were screened by in vitro adhesion method. In vitro and in vivo invasion and metastasis of sub-strains were observed by Boyden chamber method and nude mice inoculation method ability. Results: After repeated in vitro adhesion by laminin adhesion, MKN-45Lm + cell subline with high adhesion and MKN-45Lm-cell subline with low adhesion were obtained. In vitro invasion, migration and migration of Lm + cells to Matrigel were higher in Lm- cells than in Lm- cells, with high metastasis and aggressiveness. Conclusion: In vitro adhesion method can successfully screen gastric cancer cell sub-strains of specific phenotype. The adhesion of gastric cancer cells is related to invasion and metastasis.
