目的:观察搜风祛痰中药复方稳斑汤对ApoE基因敲除小鼠动脉粥样硬化易损斑块Beclin-1和Bcl-2基因表达的影响。方法:建立ApoE基因敲除小鼠动脉粥样硬化不稳定斑块模型,并设立空白组、模型组、西药组、稳斑汤低、中、高剂量组。13周后将小鼠麻醉处死,取经HE染色的主动脉组织在光学显微镜下进行病理学观察,并采用半定量RT-PCR技术检测Beclin-1和Bcl-2基因表达的变化。结果:与空白组相比,模型组Beclin-1mRNA表达水平明显下降(P<0.01);与模型组相比,西药组和稳斑汤各剂量组均能提高Beclin-1mRNA表达水平(P<0.01);与西药组相比,中药高剂量组Beclin1mRNA表达水平无明显差异(P>0.05)。与空白组对比,模型组Bcl-2mRNA表达水平显著降低(P<0.05);与模型组相比,西药组和稳斑汤各剂量组均能提高Bcl-2mRNA表达水平(P<0.01);与西药组相比,稳斑汤高剂量组Bcl-2mRNA表达水平无明显差异(P>0.05)。光镜下观察西药组和稳斑汤各剂量组炎症反应均明显弱于模型组。结论:搜风祛痰中药复方稳斑汤可能通过上调ApoE基因敲除小鼠AS易损斑块中Beclin-1和Bcl-2的基因表达而干预AS不稳定斑块的形成及破裂,具有良好的心血管保护作用。
OBJECTIVE: To observe the effect of Fengfeng and Sputum elimination traditional Chinese medicine Wenxue Tang on Beclin-1 and Bcl-2 gene expression in atherosclerotic vulnerable plaque of ApoE knockout mice. Methods: ApoE gene knockout mice model of atherosclerotic plaque was established, and blank, model group, western medicine group, Wenxue Tang low, medium and high dose groups were established. Thirteen weeks later, the mice were anesthetized and sacrificed. The aortas stained with HE were observed under a light microscope and the changes of Beclin-1 and Bcl-2 gene expression were detected by semi-quantitative RT-PCR. Results: Compared with the blank group, Beclin-1mRNA expression level of the model group was significantly decreased (P <0.01); Compared with the model group, ). Compared with Western medicine group, there was no significant difference in the expression level of Beclin1mRNA in high-dose group (P> 0.05). Compared with the blank group, the expression of Bcl-2mRNA in the model group decreased significantly (P <0.05). Compared with the model group, the western medicine group and the Wenwei Tang group could increase the expression of Bcl-2mRNA (P <0.01) Compared with western medicine group, there was no significant difference in Bcl-2mRNA expression between high-dose and high-dose group (P> 0.05). Under the light microscope, the inflammatory reaction in the western medicine group and the Wenwei Tang group were weaker than the model group. Conclusion: Fengfeng expectorant traditional Chinese medicine Wenban Decoction may interfere with the formation and rupture of AS unstable plaques by up-regulating gene expression of Beclin-1 and Bcl-2 in AS vulnerable plaque of ApoE knockout mice, which is good Cardiovascular protection.
