目的:建立保健食品及中药制剂中化学降糖药格列本脲和格列美脲的HPLC测定方法。方法:用甲醇超声提取保健食品及中药制剂中的格列本脲和格列美脲,采用高效液相色谱二级管阵列检测器进行检测,外标法定量。结果:格列本脲低浓度平均回收率(n=6)为101.7%,RSD为0.43%;中浓度为94.2%,RSD为0.48%。格列美脲低浓度平均回收率(n=6)为104.9%,RSD为1.49%;中浓度为100.7%,RSD为1.64%。格列本脲、格列美脲的方法检出限分别为0.015、0.015μg。线性范围为1.5~100 mg/L。结论:本方法操作简便,分离度好,可用于保健食品和中药制剂中格列本脲、格列美脲的定量检测。
Objective: To establish a method for the determination of glyburide and glimepiride, which are chemical hypoglycemic agents in health food and traditional Chinese medicine. Methods: Glibenclamide and glimepiride in health food and Chinese medicine preparations were extracted with methanol and detected by high performance liquid chromatography (HPLC) diode array detector with external standard method. Results: The average recovery of glyburide at a low concentration (n = 6) was 101.7% with a RSD of 0.43%, a median concentration of 94.2% and a RSD of 0.48%. The average recovery of glimepiride at low concentration (n = 6) was 104.9% with a RSD of 1.49%; the median concentration was 100.7% and the RSD was 1.64%. The detection limits of glyburide and glimepiride were 0.015 and 0.015 μg, respectively. The linear range is 1.5 to 100 mg / L. Conclusion: The method is simple and convenient to operate and has good separation. It can be used for the quantitative detection of glibenclamide and glimepiride in health food and traditional Chinese medicine.
