目的探讨趋化因子γ干扰素诱导的单核因子(Mig)、γ干扰素诱导蛋白10(IP-10)在婴儿巨细胞病毒肝炎中的作用及机制。方法2006年11月至2007年10月,武汉市儿童医院采用实时荧光定量逆转录聚合酶链反应(RT-PCR)检测39例婴儿巨细胞病毒肝炎患儿及20名健康婴儿外周血单个核细胞(PBMC)中Mig、IP-10 mRNA表达,双抗体夹心ELISA法检测血清中的Mig、IP-10水平。结果婴儿巨细胞病毒肝炎患儿PBMC中Mig、IP-10 mRNA表达水平及血清中Mig、IP-10水平均较正常对照组升高(t分别=14.860、11.232、15.511、12.800,均P<0.01)。患儿组血清Mig、IP-10水平与血清ALT呈正相关(r分别=0.6376、0.7239,均P<0.05)。结论Mig、IP-10参与了婴儿巨细胞病毒肝炎的发生过程,与疾病的严重程度有关。
Objective To investigate the role and mechanism of Mig and IFN-γ10 induced by chemokine interferon-γ in infants with cytomegalovirus hepatitis. Methods From November 2006 to October 2007, Wuhan Children’s Hospital detected 39 peripheral blood mononuclear cells from infants with cytomegalovirus and 20 healthy infants by real-time fluorescent quantitative reverse transcription-polymerase chain reaction (RT-PCR) (PBMC) Mig, IP-10 mRNA expression, double antibody sandwich ELISA method for the detection of Mig, IP-10 levels in serum. Results The levels of Mig, IP-10 mRNA and the levels of Mig and IP-10 in PBMC of infants with cytomegalovirus were significantly higher than those of the control group (t = 14.860, 11.232, 15.511 and 12.800, all P <0.01 ). Serum levels of Mig and IP-10 in children were positively correlated with serum ALT (r = 0.6376,0.7239, all P <0.05). Conclusion Mig and IP-10 are involved in the pathogenesis of infant cytomegalovirus hepatitis, which is related to the severity of the disease.
