目的 :观察丙氧鸟苷 (GCV)对A5 49-TK细胞的作用与诱导细胞凋亡的关系。方法 :A5 49-TK、A5 49两种细胞经 5 0 μmol/LGCV作用 3~ 5d后 ,用电镜、流式细胞仪、细胞凋亡原位检测法 (Tunel)观察细胞凋亡。结果 :电镜下发现A5 49-TK细胞有凋亡特征 :如凋亡小体、半月征、核浓缩 ,流式细胞仪发现A5 49-TK细胞有亚G0 G1峰 ,凋亡细胞占 12 2 1%± 1 6 6 % ,而A5 49细胞无G0 G1峰 ,凋亡细胞占 1 32 %±0 2 5 % ,两者比较P <0 0 1。Tunel表明A5 49-TK、A5 49两种细胞凋亡发生率分别为 16 %± 1 6 7% ,2 5 %±0 36 % ,两者比较P <0 0 1。结论 :GCV诱导A5 49-TK细胞凋亡
Objective: To observe the effect of gonadoside (GCV) on A549-TK cells and its relationship with apoptosis. Methods: A549-TK and A549 cells were treated with 50 μmol / L LGC for 3 ~ 5 days. Cell apoptosis was observed by electron microscopy, flow cytometry and in situ cell death assay (Tunel). Results: The apoptosis of A549-TK cells was observed under electron microscope: A549-TK cells were found to have sub-G0 G1 peak by flow cytometry and apoptotic cells accounted for 12 2 1 % ± 1 6 6%, while A549 cells had no G0 G1 peak and apoptotic cells accounted for 132% ± 0 25%, both P <0.01. Tunel showed that the rates of apoptosis of A549-TK and A549 cells were 16% ± 1 67% and 25% ± 0 36%, respectively, P <0.01. Conclusion: GCV induced the apoptosis of A549-TK cells
