目的通过实验观察绿茶提取物—茶色素(TeaPigment,TP)对豚鼠心肌氧自由基损伤的保护作用。方法取豚鼠右室前乳头肌,置于Tyrode液33±1℃浴槽中,通95%O2和5%CO2混合气体。1组加黄嘌呤—黄嘌呤氧化酶系统(XXOD)产生氧自由基诱发乳头肌氧化损伤;另1组在加XXOD之前5min先加入TP25mg/L。分别观察各组心室乳头肌的生理特性变化,最后测定各标本匀浆中超氧化歧化酶(SOD)和脂质过氧化物丙二醛(MDA)的含量。结果TP能显著对抗氧自由基损伤诱导的心肌细胞膜损伤和心肌生理特性的变化,对心肌组织中SOD活性降低及MDA含量升高,有明显对抗作用。结论TP对心肌细胞膜氧自由基损伤有显著的保护作用
Objective To observe the protective effect of green tea extract (TP) on myocardial oxygen free radical injury in guinea pigs. Methods The anterior papillary muscle of right ventricle of guinea pig was placed in a bath of 33 ± 1 ℃ in Tyrode solution and mixed with 95% O2 and 5% CO2. One group of xanthine-xanthine oxidase system (X XOD) produce oxygen free radicals induced papillary muscle oxidative damage; the other group before adding X XOD 5min before adding TP25mg / L. The changes of physiological characteristics of the papillary muscles in each group were observed. The content of superoxide dismutase (SOD) and lipid peroxides (MDA) in the homogenate of each sample were measured. Results TP could significantly antagonize the changes of myocardial cell membrane damage and myocardial physiological characteristics induced by oxygen free radical injury, and significantly antagonize the decrease of SOD activity and MDA content in myocardial tissue. Conclusion TP has a significant protective effect on myocardial cell oxygen free radical damage
