在单因素试验的基础上,根据中心组合试验设计原理,利用响应面分析法对桦褐孔菌总三萜超声提取过程中3个主要影响因素:超声时间、超声温度、料液比进行了优化。结果表明,超声提取温度、时间对三萜得率影响最为显著(p<0.001),最佳工艺条件为:采用异丙醇做提取溶剂,料液比为23mL/g,在60℃下超声提取22min。在此最佳条件下,总三萜得率为5.40%,试验结果与模型预测值相符。
On the basis of single factor experiment, according to the principle of central composite experimental design, response surface analysis was used to optimize three major influencing factors of ultrasonic extraction time, ultrasonic temperature and solid-liquid ratio . The results showed that ultrasonic extraction temperature and time had the most significant effect on the yield of triterpene (p <0.001). The optimum conditions were as follows: extraction solvent was isopropanol, the ratio of solid to liquid was 23mL / g, ultrasonic extraction was carried out at 60 ℃ 22min. Under this optimum condition, the total triterpene yield was 5.40%, and the test results were in agreement with the model predictions.
