目的 识别确认中国人群的HLA新等位基因。方法 经基因克隆和DNA测序 ,分析该基因和B 5 6 0 2基因序列的差异。结果 该基因和B 5 6 0 2基因序列的差异只是在外显子 3区域中 5 5 9C >A以及 5 6 0T >C 2个碱基取代 ,并导致相应的密码子 187由亮氨酸变为苏氨酸。结论 该基因为HLA新等位基因 ,2 0 0 2年 9月已被世界卫生组织HLA因子命名委员会正式命名为HLA B 5 6 10。在大约 30 0 0名随机献血者中 ,未发现其他带有B 5 6 10基因的个体。
Objective To identify and identify new HLA alleles in Chinese population. Methods The gene and B 5 6 0 2 gene sequences were analyzed by gene cloning and DNA sequencing. As a result, the difference between this gene and the B 5 6 0 2 gene sequence was only 5 5 9 C> A and 5 6 0 T> C 2 base substitutions in the exon 3 region and the corresponding codon 187 was changed from leucine to Threonine. Conclusion This gene is a new allele of HLA. In September 2002, it was officially named as HLA B 5 6 10 by the WHO Nomenclature Committee of HLA. Among approximately 300 randomized donors, no other individuals with the B 5 6 10 gene were found.
