目的:在原代培养的小鼠肝细胞上观察白细胞介素1β(IL1β)对醋氨酚诱导的细胞损伤的影响及其与一氧化氮、环磷酸鸟苷(cGMP)的关系。方法:以两步灌流法分离小鼠肝细胞,进行原代培养;以放射免疫法测定细胞内cGMP和环磷酸腺苷(cAMP)含量。结果:IL1β可减轻醋氨酚诱导的转氨酶漏出,增加肝细胞内cGMP含量,一氧化氮合酶阻断剂LNMMA可完全阻断IL1β的抗损伤作用。结论:IL1β对醋氨酚诱导的肝细胞损伤具有直接的保护作用,这一作用可能是通过一氧化氮、cGMP介导的。
OBJECTIVE: To observe the effects of interleukin-1β (IL1β) on acetaminophen-induced cell injury in primary cultured mouse hepatocytes and its relationship with nitric oxide and cGMP. Methods: Mouse hepatocytes were isolated by two-step perfusion method and cultured in primary culture. The contents of cGMP and cAMP in cells were determined by radioimmunoassay. Results: IL 1β can reduce paracetamol-induced transaminase leakage, increase the content of cGMP in liver cells, nitric oxide synthase inhibitor L NMMA can completely block the IL 1β anti-injury effect. Conclusion: IL 1β has a direct protective effect against acetaminophen-induced hepatocellular injury, which may be mediated by nitric oxide and cGMP.
