目的 探讨慢性铅暴露对小鼠脑海马神经元蛋白激酶B表达的影响。方法 5~6周龄小鼠交配后,通过饮水饲以终浓度(0,2.4,4.8,9.6mmol·L~-1)醋酸铅制备动物模型。用Western blot法观察各组小鼠脑海马神经元总量PKB(t-PKB)及磷酸化PKB(p-PKB)的表达变化。结果 染铅组小鼠血铅和脑铅浓度明显升高,与对照组比较有统计学意义(p<0.01)。染铅组脑海马神经元p-PKB表达水平比对照组明显降低,并与血铅浓度有效应关系,呈负相关(r=-0.84,p<0.01)。铅对小鼠大脑海马神经元细胞t-PKB的表达水平没有影响。结论 慢性铅暴露导致小鼠脑海马神经元中p-PKB表达的改变可能与小鼠学习记忆功能减退有关。
Objective To investigate the effects of chronic lead exposure on the expression of protein kinase B in brain hippocampal neurons in mice. Methods Mice aged 5 to 6 weeks were mated and fed with drinking water to prepare animal model of lead (0,2.4,4.8,9.6 mmol·L -1) lead acetate. Western blot was used to observe the changes of the total amount of PKB (t-PKB) and phosphorylated PKB (p-PKB) in the hippocampal neurons of each group. Results The levels of lead and lead in blood of lead-exposed mice were significantly increased compared with the control group (p <0.01). The expression of p-PKB in the lead-exposed group was significantly lower than that in the control group, and negatively correlated with the effective level of blood lead (r = -0.84, p <0.01). Lead had no effect on the level of t-PKB in hippocampal neurons of mice. Conclusion The change of p-PKB expression in the hippocampal neurons of mice induced by chronic lead exposure may be related to the decline of learning and memory in mice.
