[目的]探讨肝细胞性肝癌(hepatocellular carcinoma,HCC)中与硫酸乙酰肝素糖蛋白3(glypican3,GPC3)表达相关的Wnt信号通路及其他信号转导分子表达。[方法]通过实时荧光定量PCR方法检测GPC3过表达的肝癌细胞株以及不同肝病变组织样本中GPC3、Wnt、基质金属蛋白酶(MMPs)、内皮硫酸脂酶及其他信号分子表达。[结果]肝癌组织GPC3mRNA水平显着高于良性肝病变组织(P<0.05)。肝癌GPC3的表达与MMP2、14,内皮硫酸脂酶1、2,成纤维细胞生长因子及其受体FGF2、FGFR1,以及胰岛素样生长因子受体IGFR1R的表达具相关性。[结论]GPC3与MMPs、FGF和IGF结合,可能在肝癌的发展中起重要作用。
[Objective] To investigate the Wnt signaling pathway and other signal transduction molecules involved in the expression of glypican3 (GPC3) in hepatocellular carcinoma (HCC). [Method] The expressions of GPC3, Wnt, matrix metalloproteinases (MMPs), endothelin and other signaling molecules in GPC3 overexpression hepatocellular carcinoma cell lines and different liver lesions were detected by real-time fluorescence quantitative PCR. [Results] The GPC3 mRNA level in HCC tissues was significantly higher than that in benign liver lesions (P <0.05). The expression of GPC3 in hepatocellular carcinoma was correlated with the expression of MMP2, 14, endothelin 1,2, fibroblast growth factor and its receptors FGF2, FGFR1, and insulin-like growth factor receptor IGFR1R. [Conclusion] GPC3 binding to MMPs, FGF and IGF may play an important role in the development of liver cancer.
