目的肥胖呈现全球流行趋势。目前,已基本证实其为一种多基因遗传病,先前研究小组筛选获得肥胖相关的新基因6次跨膜表位抗原(six-transmembrane epithelial antigen of the prostate 4,STEAP4)在肥胖患儿脂肪组织中差异表达,旨在进一步验证其在肥胖儿童脂肪组织中的表达,并初步探讨STEAP4的生物信息学特征。方法采用实时荧光定量PCR(Realtime-qPCR)技术验证肥胖与健康儿童脂肪组织中STEAP4基因的表达差异,应用DNA Star、Blast、ProtParam、SOPMA、Tar-getP、TMHMM、ProtScale、Interpro Scan和SMART等软件或数据库分析STEAP4的核苷酸基本特征、蛋白质理化性质、信号肽、跨膜结构域、疏水性/亲水性、结构域及亚细胞定位等。结果 STEAP4基因是一个功能未知的基因,低表达于肥胖患儿脂肪组织中,生物信息学分析该基因cDNA全长4454 bp,开放阅读框(open reading frame,ORF)长1380 bp,编码459个氨基酸,预测相对分子质量51981.2,定位在胞膜,蛋白序列分析显示为6次跨膜螺旋结构,为可溶的亲水性蛋白,蛋白结构域分析显示具有黄素蛋白跨膜成分(三铁还原酶结构域),NAD(P)结合结构域,NADP氧化还原酶(依赖辅酶F420)和血红素蛋白保守位点。结论 STEAP4基因是一低表达于肥胖患儿脂肪组织中的基因,高度提示该蛋白与肥胖及其相关的胰岛素抵抗的发生存在相关性,值得进一步研究证实。
Purpose Obesity presents a global trend. At present, it has been basically confirmed that it is a polygenic disease. Previously, the research team screened the obesity-related new gene 6-transmembrane epithelial antigen of the prostate 4 (STEAP4) in obese children with adipose tissue In order to further verify its expression in adipose tissue of obese children and to explore the bioinformatics characteristics of STEAP4. Methods Real-time quantitative PCR (Realtime-qPCR) was used to validate the expression of STEAP4 gene in adipose tissues of obese and healthy children. The softwares of DNA Star, Blast, ProtParam, SOPMA, Tar-getP, TMHMM, ProtScale, Interpro Scan and SMART Or database analysis of basic nucleotide nucleotide characteristics of STEAP4, protein physicochemical properties, signal peptide, transmembrane domain, hydrophobic / hydrophilic, domain and subcellular localization. Results STEAP4 gene was a gene of unknown function and was overexpressed in adipose tissue of obese children. Bioinformatics analysis revealed that the full-length cDNA of STEAP4 gene was 4454 bp in length and the open reading frame (ORF) was 1380 bp in length, encoding 459 amino acids , The predicted molecular mass of 51981.2, located in the membrane, protein sequence analysis showed a 6-transmembrane helix structure as a soluble hydrophilic protein, protein domain analysis showed that the membrane element of flavoprotein (ferriferous reductase Domain), NAD (P) binding domain, NADP oxidoreductase (coenzyme F420 dependent), and heme protein conserved sites. Conclusion The STEAP4 gene is a low expression in adipose tissue of obese children, suggesting that there is a correlation between STEAP4 gene and obesity and its related insulin resistance. It is worth further study to confirm.
