目的:观察黔棘茎楤木水煎液对大鼠佐剂性关节炎(AA)的作用,并探讨其可能的作用机制。方法:60只Wistar大鼠随机分为正常组、模型组、雷公藤多苷片组(12 mg/kg)、黔棘茎楤木低、中、高剂量组(0.88、1.75、3.5 g/kg);除正常组外,其余各组均采用弗氏完全佐剂足跖皮下注射复制AA模型,给药组大鼠连续35d灌胃给药,正常组、模型组予等容量的生理盐水。给药结束后,X线拍摄左后足踝关节侧位片,采用x硝酸盐还原法检测各组大鼠血清中踝关节局部前列腺素E2(PGE2)含量。结果:模型组大鼠踝关节面明显毛糙,关节间隙变窄、消失,踝关节局部的PGE2含量均较正常组明显增加(P<0.01);与模型组比较雷公藤多苷片组、黔棘茎楤木高、中剂量组均明显降低了踝关节局部的PGE2含量(P<0.01,P<0.05);与雷公藤多苷片组比较,黔棘茎楤木高剂量组踝关节局部的PGE2含量无显著差异(P﹥0.05)。结论:黔棘茎楤木对AA大鼠关节有明显治疗作用,其机制与抑制炎性细胞因子或炎症介质有关。
Objective: To observe the effect of Qianji Stemwood decoction on adjuvant arthritis (AA) in rats and its possible mechanism. Methods: Sixty Wistar rats were randomly divided into three groups: normal group, model group, tripterygium glycosides tablet (12 mg / kg), low, medium and high dose ). Except for the normal group, AA rats were subcutaneously injected with complete Freund’s adjuvant footpad. The rats in the treated group were given gavage for 35 days continuously, and the rats in the normal group and normal saline group were given the same volume of normal saline. At the end of the administration, X-ray was taken to record the left lateral ankle and the X-ray was used to detect the content of prostaglandin E2 (PGE2) in the ankle joint of rats in each group by x nitrate reduction. Results: The ankle joint surface of the model group was obviously rough and the joint space narrowed and disappeared. The content of PGE2 in the ankle joint area of the model group was significantly higher than that of the normal group (P <0.01). Compared with the model group, Compared with tripterygium wilfordii tablet group, the PGE2 content in the ankle joint of high-dose and low-dose group was significantly decreased (P <0.01, P <0.05) There was no significant difference (P> 0.05). Conclusion: The stem of Alnus cremastogyne has a significant therapeutic effect on the joints of AA rats, and its mechanism is related to the inhibition of inflammatory cytokines or inflammatory mediators.
