目的观察海风藤提取物灌胃对高尿酸血症小鼠血清尿酸水平的影响及对大鼠痛风性炎症的防治作用。方法取雄性昆明种小鼠100只随机分为A、B组各50只。A、B组各分为5个亚组,其中3个亚组分别以高(9.00 g/kg)、中(4.50 g/kg)、低(2.25 g/kg)剂量的海风藤提取物灌胃,另2个亚组分别给予20 mg/kg别嘌醇片和等体积蒸馏水灌胃,A、B两组灌胃均1次/d、连续12 d。A组于末次灌胃后1 h,腹腔注射次黄嘌呤600 mg/kg制作高尿酸血症模型,B组不给予次黄嘌呤。A、B组分别于腹腔注射后2.5 h、末次灌胃1 h后眼眶静脉取血,采用磷钨酸比色法检测血清尿酸。取雄性Wistar大鼠40只随机分为1、2、3、4组各10只。1、2组分别采用高(9.00 g/kg)、低(2.25 g/kg)剂量的海风藤提取物灌胃,3、4组予等体积蒸馏水灌胃,各组灌胃1次/d,连续10 d。第10天灌胃1h后取1、2、3组大鼠于右后足跖皮下注射100 g/L的尿酸钠结晶0.15 m L诱导痛风性炎症。分别于尿酸钠结晶注射后1、2、3、4、5、6 h观察各组大鼠右后足跖肿胀情况,计算肿胀度。结果 A组的高剂量海风藤提取物、中剂量海风藤提取物亚组小鼠血清尿酸水平低于A、B组的蒸馏水亚组(P均<0.05)。3组大鼠尿酸钠结晶注射后不同时间右后足跖肿胀度均高于4组(P均<0.05)。尿酸钠结晶注射后3、4、6 h时1组大鼠右后足跖肿胀度低于3组(P均<0.05)。尿酸钠结晶注射后2 h时2组大鼠右后足跖肿胀度低于3组(P<0.05)。结论海风藤提取物可降低高尿酸血症小鼠血清尿酸水平,防治大鼠痛风性炎症。
Objective To observe the effects of Piper kadsura Gavage on serum uric acid level in hyperuricemic mice and its preventive and therapeutic effects on gouty inflammation in rats. Methods 100 male Kunming mice were randomly divided into A and B groups of 50 rats. On the 10th day after gavage, rats in Groups 1 and 2 were given gavage inflammation by subcutaneously injecting 0.15 g of 100 g / L sodium urate into the right posterior plantar. The swelling of the right hind paw of each group was observed at 1, 2, 3, 4, 5 and 6 h after sodium urate crystal injection, and the degree of swelling was calculated. Results The level of serum uric acid in group A was lower than that in group A and group B (all P <0.05). The swelling degree of right hind paw at different time after sodium urate crystal injection in 3 groups was higher than that in 4 groups (all P <0.05). The swelling of the right hind paw in 1 group was lower than that in 3 groups 3, 4 and 6 h after sodium urate crystal injection (all P <0.05). At 2 h after sodium urate injection, the swelling of the right hind paw in both groups was lower than that in the 3 groups (P <0.05). Conclusion Piper kadsura extract can reduce serum uric acid in hyperuricemic mice and prevent and cure gout inflammation in rats.
