目的考察替丁类药物与全肠外营养液(total parenteral nutrient,TPN)配伍后的体外稳定性及抗溃疡作用。方法根据临床处方配比,将替丁类药物加入到全肠外营养液中,得到不同浓度的含药制剂及对照制剂,配伍液在室温下放置24h,于不同时间点观察、测定和记录配伍液的外观性状、脂肪乳粒径大小及分布、pH值和渗透浓度;建立组胺致小鼠溃疡模型,所得数据采用SPSS统计分析软件进行t检验,观察西咪替丁(cimetidine,CMT)、法莫替丁(famotidine,FMT)与TPN配伍制剂对胃溃疡的抑制作用。结果替丁类药物与全肠外营养液配伍后,制剂的外观、脂肪乳粒径大小及分布、pH值和渗透浓度在24h内均没有发生显著性变化;药效学实验中,CMT-TPN(50.0、100.0 mg/kg)和FMT-TPN(4.2、8.4mg/kg)均可显著抑制组胺致小鼠溃疡的发生,其溃疡抑制率分别为49.44%、60.69%和53.52%、67.29%。结论替丁类药物与TPN配伍后能够稳定24h,且有显著预防和治疗胃溃疡的作用,本研究为其临床配伍使用提供了数据支持。
Objective To investigate the in vitro stability and anti-ulcer effect of steroids combined with total parenteral nutrition (TPN). In the pharmacodynamic experiments, CMT-TPN (50.0, 100.0 mg / kg) and FMT-TPN (4.2, 8.4 mg / kg) significantly inhibited the histamine-induced ulcer in mice. The ulcer inhibition rates were 49.44%, 60.69% and 53.52%, 67.29% . Conclusion The drug combination with TPN can stabilize for 24h and has a significant effect in preventing and treating gastric ulcer. This study provides data support for its clinical compatibility.
