目的探讨ErbB3和Flotillin-2在骨肉瘤中的表达及其相关性,并分析其临床意义。方法取2009年9月-2014年3月收治的38例骨肉瘤患者和13例骨软骨瘤患者样本组织,行免疫组织化学染色。镜下观察ErbB3和Flotillin-2的表达情况,并统计分析二者在骨肉瘤和骨软骨瘤中阳性表达差异性、二者相关性及其与临床特征的关系。结果镜下观察示,ErbB3、Flotillin-2蛋白均在骨肉瘤组织细胞质中高表达,其阳性率显著高于骨软骨瘤组织中(P<0.05)。ErbB3和Flotillin-2在骨肉瘤中的表达具有较好一致性(Kappa值=0.434,P=0.000)。ErbB3在骨肉瘤中表达与患者性别、年龄、肿瘤部位、肿瘤大小不相关(P>0.05),与美国国立综合癌症网络(NCCN)指南骨肉瘤临床分期及肺转移显著相关(P<0.05);而Flotillin-2在骨肉瘤中的表达与以上因素均无相关(P>0.05)。影响骨肉瘤患者总生存期的因素包括骨肉瘤临床分期、肺转移、ErbB3表达阳性及肿瘤大小(P<0.05),独立影响因素是肺转移和ErbB3表达阳性(P<0.05)。结论 ErbB3和Flotillin-2在骨肉瘤中过表达,且具有一致性。此外,ErbB3的表达与骨肉瘤临床分期和肺转移有关,肺转移和ErbB3过表达提示预后不佳。
Objective To investigate the expression and correlation of ErbB3 and Flotillin-2 in osteosarcoma and to analyze its clinical significance. Methods Thirty-eight osteosarcoma patients and 13 osteochondroma samples from September 2009 to March 2014 were collected and immunohistochemically stained. The expression of ErbB3 and Flotillin-2 was observed microscopically. The differences of positive expression between ErbB3 and Flotillin-2 in osteosarcoma and osteochondroma were statistically analyzed, and the correlation between ErbB3 and Flotillin-2 was also analyzed. Results The results of microscopic examination showed that both ErbB3 and Flotillin-2 protein were highly expressed in the cytoplasm of osteosarcoma and the positive rate was significantly higher than that in osteochondroma (P <0.05). The expression of ErbB3 and Flotillin-2 in osteosarcoma showed good agreement (Kappa = 0.434, P = 0.000). The expression of ErbB3 in osteosarcoma was not correlated with patient ’s sex, age, tumor location and tumor size (P> 0.05), but was significantly correlated with the clinical stage and lung metastasis of osteosarcoma (NCCN) according to the guidelines of American National Comprehensive Cancer Network (NCCN). The expression of Flotillin-2 in osteosarcoma had no correlation with the above factors (P> 0.05). The factors affecting the overall survival of patients with osteosarcoma include osteosarcoma clinical stage, lung metastasis, ErbB3 positive expression and tumor size (P <0.05). The independent influencing factors were lung metastasis and ErbB3 positive expression (P <0.05). Conclusion ErbB3 and Flotillin-2 are overexpressed and consistent in osteosarcoma. In addition, the expression of ErbB3 is associated with clinical stage and lung metastasis of osteosarcoma. Lung metastasis and ErbB3 overexpression indicate poor prognosis.
