目的了解转铁蛋白受体(transferrin receptor,TfR)在皮肤黑素瘤中的水平及其临床意义。方法采用免疫组化法检测34例皮肤黑素瘤和20例色素痣石蜡标本中TfR的水平,流式细胞仪(FCM)检测TfR在黑素瘤细胞系A375,A875和KZ28细胞株的表达,并以文献报道的TfR阴性表达的人卵巢癌细胞系A2780作为阴性对照。结果在黑素瘤组织中,TfR的水平较色素痣对照组的水平明显增加,差异有统计学意义(P<0.05),并且TfR的水平与黑素瘤的病理分期、clark分级和有丝分裂均具有相关性;在黑素瘤细胞水平上,TfR在黑素瘤细胞系均高水平,且以A375水平最高。结论 TfR在皮肤黑素瘤中的水平明显增高,这为未来使用TfR作为基因治疗黑素瘤的靶向分子提供了坚实理论依据。
Objective To investigate the level of transferrin receptor (TfR) in cutaneous melanoma and its clinical significance. Methods Immunohistochemistry was used to detect TfR levels in 34 cases of melanoma and 20 cases of nevus. The expression of TfR in melanoma cell lines A375, A875 and KZ28 was detected by flow cytometry (FCM) A negative TfR-expressing human ovarian cancer cell line A2780 was also reported as a negative control. Results The level of TfR in melanoma tissue was significantly higher than that in control group (P <0.05), and the level of TfR was correlated with the pathological stage of melanoma, clark grading and mitosis Correlation; at melanoma cell level, TfR was high in melanoma cell lines and highest at A375 level. Conclusion The level of TfR in cutaneous melanoma is significantly increased, which provides a solid theoretical basis for the future use of TfR as a target molecule for gene therapy of melanoma.
