目的初步研究腺样囊性癌细胞系(SACC83)与肺高转移的腺样囊性癌细胞系(SACC-LM)中MMP-1、MMP-2、MMP-3和MMP-7的表达差异。方法通过免疫细胞化学法、RT-PCR、Western blot研究不同转移特性的腺样囊性癌细胞系SACC-LM、SACC83中基质金属蛋白酶(MMP-1、MMP-2、MMP-3、MMP-7)在mRNA和蛋白质水平的表达。结果免疫细胞化学法中MMP-1、-2、-3、-7在两种腺样囊性癌细胞均表达。在mRNA水平,MMP-1、-2、-3、-7在两种腺样囊性癌细胞的表达差异无显著性统计学意义。但是在Western blot实验中,SACCLM中MMP-1、-2、-3的蛋白质相对表达量较SACC83增多,差异有统计学意义。结论基质金属蛋白酶MMP-1、-2、-3在肺高转移特性的腺样囊性癌细胞系表达增高对于研究腺样囊性癌血行转移的分子机制具有一定的意义。
Objective To study the differential expression of MMP-1, MMP-2, MMP-3 and MMP-7 in adenoid cystic carcinoma cell line (SACC83) and highly metastatic adenoid cystic carcinoma cell line (SACC-LM) Methods The expressions of MMP-1, MMP-2, MMP-3 and MMP-7 in adenoid cystic carcinoma cell lines SACC-LM and SACC83 with different metastatic characteristics were studied by immunocytochemistry, RT-PCR and Western blot ) At mRNA and protein levels. Results Immunocytochemistry showed that MMP-1, -2, -3 and -7 were expressed in both adenoid cystic carcinoma cells. There was no significant difference in the expression of MMP-1, -2, -3, -7 between the two kinds of adenoid cystic carcinoma cells at mRNA level. However, in Western blot, the relative expression of MMP-1, -2, -3 in SACCLM protein increased compared with SACC83, the difference was statistically significant. Conclusion The increased expression of matrix metalloproteinase MMP-1, -2, -3 in adenoid cystic carcinoma cell line with high lung metastasis is of great significance for studying the molecular mechanism of hematogenous metastasis of adenoid cystic carcinoma.
