RAPD molecular markers and ITS sequence comparisons were used to establish the relationship between Boletus appendiculatus fruit bodies collected from Qingyuan, Zhejiang Province, and mycelium isolated from the fruit body tissue. RAPD analysis of extracted genomic DNA revealed that B. appendiculatus fruit bodies (fb) and the isolated fungal mycelium (mb) formed a single cluster with a Dice coefficient value of 0.92. Comparison of ITS sequences from these samples showed that base sequences and fragment lengths were identical, thereby confirming that the fungal mycelium isolated from B. appendiculatus fruit bodies was the vegetative form of the mushroom. Our data suggest that RAPD fingerprinting combined with ITS sequence comparisons constitute an effective and feasible method for identifying mycelium isolated from ectomycorrhizal fungi.
