目的:研究亮叶围涎树根的化学成分及其抗肿瘤活性。方法:应用硅胶,Sephadex LH-20,HPLC等各种色谱技术进行分离纯化,用NMR等谱学方法分析确定化合物结构。采用MTT法对分离得到的化合物进行抗肿瘤活性评价。结果:从亮叶围涎树根的95%乙醇提取物的乙酸乙酯萃取物中分离得到6个化合物,分别鉴定为:julibroside A2(1),3-[(2-acetamido-2-deoxy-β-D-glucopyranosyl)oxy]-16α-hydroxyolean-12-en-28-oic acid(2),没食子酸(3),没食子酸乙酯(4),(+)-儿茶素(5),(-)-没食子儿茶素没食子酸酯(6)。化合物2对人源肿瘤细胞A2780具有较强的细胞毒活性(IC501.72μmol.L-1)。结论:化合物1~6均为首次从该植物中分离获得。化合物2对人源肿瘤细胞A2780具有明显的细胞毒活性。
Objective: To study the chemical constituents and their antitumor activity of bark of bryophytes. Methods: The compounds were isolated and purified by silica gel, Sephadex LH-20, HPLC and other chromatographic techniques. The structures of the compounds were determined by NMR spectroscopy. The antitumor activity of the isolated compounds was evaluated by MTT assay. Results: Six compounds were isolated from the ethyl acetate extract of 95% ethanol extract of the leaves of Radix salivarius, identified as julibroside A2 (1), 3 - [(2-acetamido-2-deoxy- β-D-glucopyranosyl oxy] -16α-hydroxyolean-12-en-28-oic acid (2), gallic acid (3), ethyl gallate (-) - gallocatechin gallate (6). Compound 2 has strong cytotoxic activity on human tumor cell A2780 (IC501.72μmol.L-1). Conclusion: Compounds 1 ~ 6 were isolated from this plant for the first time. Compound 2 has obvious cytotoxic activity on human tumor cell A2780.
