【目的】观察益气活血代表药物人参、三七的提取物在不同比例配伍下对大鼠腹膜间皮细胞(PMCs)增殖的影响。【方法】采用胰蛋白酶消化法分离大鼠PMCs,建立稳定的细胞培养模型,加入含42.5g/L葡萄糖的腹膜透析液作用3h后,再加入不同比例配伍的人参总皂苷和三七总皂苷,采用四甲基偶氮唑盐(MTT)法检测不同浓度的药物及其配伍对大鼠PMCs增殖的影响。【结果】人参总皂苷和三七总皂苷终浓度为100μg/mL及80μg/mL均能对高糖透析液损伤后的PMCs的增殖产生保护作用(P<0.01);三七总皂苷与人参总皂苷在浓度比为1∶9以及4∶1时能改善42.5 g/L腹透液作用3 h后对细胞产生的增殖抑制(P<0.01),且两组的保护效能差异无显著性意义(P>0.05)。【结论】具有益气活血作用的人参与三七提取物在一定比例配伍后能够对高糖刺激下的大鼠PMCs产生一定的保护作用。
【Objective】 To observe the effects of the extracts of Panax ginseng and Panax notoginseng on the proliferation of rat peritoneal mesothelial cells (PMCs) under different proportions. 【Methods】 Rat PMCs were isolated by trypsin digestion and established a stable cell culture model. After adding peritoneal dialysis solution containing 42.5g / L glucose for 3h, different concentrations of ginsenoside and total saponins of Panax notoginseng were added, The effects of different concentrations of drugs and their compatibility on the proliferation of rat PMCs were detected by MTT assay. 【Results】 The results showed that the total ginsenosides and the total saponins of Panax notoginseng saponins at a concentration of 100μg / mL and 80μg / mL all could protect the proliferation of PMCs after high glucose dialysate injury (P <0.01) Saponin could improve the proliferation inhibition of cells (P <0.01) after 42.5 g / L dialysate 3 h at the concentration ratios of 1: 9 and 4: 1, and there was no significant difference between the two groups P> 0.05). 【Conclusion】 Panax ginseng extract and Panax notoginseng extract, which have the effect of supplementing qi and activating blood circulation, can protect rat PMCs stimulated by high glucose to a certain extent.
