目的研究类风湿关节炎(RA)患者体内自身反应性T细胞的抗原互补区3(CDR3)的序列特性。方法采用实时定量聚合酶链反应(PCR)的方法扩增带有BV14和BV16的自身反应性T细胞抗原受体β链,用基因扫描的方法分析抗原互补区3的片断长度,用序列测定的方法分析CDR3的氨基酸序列。结果RA患者关节滑膜浸润T细胞呈寡克隆扩增,带有BV14和BV16的自身反应性T细胞分别取用不同VDJ片段组合,自身反应性T细胞CDR3长度有15bp、21bp和24bp三种形式,在不同患者自身反应性T细胞CDR3中发现相同氨基酸序列。结论RA患者自身反应性T细胞在病灶部位受到某些抗原的驱动而发生受体取用的偏移和CDR3的特异性扩增。
Objective To study the sequence characteristics of the autoantigen T cell complementary antigen 3 (CDR3) in patients with rheumatoid arthritis (RA). Methods The real-time quantitative polymerase chain reaction (PCR) method was used to amplify the beta chain of autoreactive T cell antigen receptor with BV14 and BV16. The fragment length of antigen complementary region 3 was analyzed by gene scanning method. Methods The amino acid sequence of CDR3 was analyzed. Results The synovial infiltrating T cells of RA patients showed oligoclonal amplification. The autoreactive T cells with BV14 and BV16 were taken by different combinations of VDJ fragments. The CDR3 length of autoreactive T cells was 15bp, 21bp and 24bp , The same amino acid sequence was found in the CDR3 of autoreactive T cells of different patients. CONCLUSIONS: Autoreactive T cells in RA patients are driven by certain antigens at the site of the lesion and undergo a shift of receptor access and CDR3-specific amplification.
