目的研究Th17细胞、IL-17、IL-23和RORγt在慢性再生障碍性贫血(CAA)患者外周血中的表达和意义。方法应用流式细胞术(FCM)检测30例CAA患者及10例健康对照外周血Th17细胞表达比例,实时荧光定量逆转录-聚合酶链反应(RT-PCR)法检测IL-17mRNA、IL-23p19mRNA和RORγtmRNA的表达水平,ELISA法检测IL-17、IL-23的表达水平。结果 CAA患者Th17细胞、IL-17mRNA、IL-23p19mRNA、RORγtmRNA、IL-17、IL-23的表达均高于健康对照组。结论 Th17细胞、IL-17、IL-23和RORγt在CAA中异常表达,提示Th17细胞、IL-17、IL-23和RORγt在CAA发病中有一定作用。
Objective To investigate the expression and significance of Th17 cells, IL-17, IL-23 and RORγt in peripheral blood of patients with chronic aplastic anemia (CAA). Methods The expression of Th17 cells in peripheral blood of 30 patients with CAA and 10 healthy controls were detected by flow cytometry (FCM). The expression of IL-17 mRNA and IL-23p19 mRNA were detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction (RT-PCR) And RORγtmRNA expression levels, ELISA detection of IL-17, IL-23 expression levels. Results The expression of Th17 cells, IL-17 mRNA, IL-23p19 mRNA, RORγt mRNA, IL-17 and IL-23 in CAA patients were higher than those in healthy controls. Conclusion Th17 cells, IL-17, IL-23 and RORγt are abnormally expressed in CAA, suggesting that Th17 cells, IL-17, IL-23 and RORγt may play a role in the pathogenesis of CAA.
