目的探讨VERGE蛋白在小鼠大脑中动脉梗死模型(MCAO)中蛋白表达的变化及其机制。方法C57BL/6小鼠30只,随机分为2组:30 min MCAO模型组、90 min MCAO模型组,制备MCAO模型,应用激光多普勒血流仪进行术中脑血流监测,应用Western印迹法观察各组梗死侧及非梗死侧的VERGE、p-JNK2、p-ERK1/2、P-38蛋白表达的变化。结果30 min、90 min MCAO模型组梗死侧VERGE、p-JNK2的蛋白表达明显增高,p-ERK1/2、P-38蛋白表达无变化;两组MCAO模型非梗死侧的VERGE、p-JNK2、p-ERK1/2、P-38蛋白表达均无变化。结论VERGE蛋白表达在小鼠MCAO的早期明显升高可能与JNK2通路有关而与ERK、P38通路无关。
Objective To investigate the changes of protein expression of VERGE protein in mouse middle cerebral artery occlusion model (MCAO) and its mechanism. Methods Thirty C57BL / 6 mice were randomly divided into 2 groups: 30 min MCAO model group and 90 min MCAO model group. The MCAO model was established. Cerebral blood flow was monitored by laser Doppler flowmetry. Western blot Methods The changes of VERGE, p-JNK2, p-ERK1 / 2 and P-38 protein expression in infarcted and non-infarcted sides of each group were observed. Results The protein expression of VERGE and p-JNK2 at infarction side of MCAO model group was significantly increased at 30 min and 90 min, while the expression of p-ERK1 / 2 and P-38 protein remained unchanged. The VERGE, p-JNK2, There was no change in p-ERK1 / 2 and P-38 protein expression. Conclusion VERGE protein expression in early stage of mouse MCAO may be related to JNK2 pathway but not to ERK and P38 pathway.
