目的:观察活血通督汤对脊髓损伤后肢体运动功能和胶质瘢痕形成的影响,探讨活血通督汤治疗脊髓损伤的作用机制。方法:成年SD大鼠18只,采用随机数表法随机分为假手术组、模型组和活血通督汤组,每组各6只。假手术组仅行椎板切除术,模型组和活血通督汤组采用NYU脊髓打击器建立脊髓损伤模型,造模后假手术组和模型组给予生理盐水灌胃,活血通督汤组给予活血通督汤灌胃。术后第1,3,5,7天行BBB肢体运动功能评分,7d后取材。采用尼氏染色法观察神经元细胞形态结构以及神经元受损情况,免疫荧光检测胶质纤维酸性蛋白(GFAP)表达,计算GFAP阳性细胞数。结果:自术后第3天起,活血通督汤组的大鼠BBB评分高于模型组大鼠BBB评分,差异有统计学意义(P<0.05);尼氏染色结果显示活血通督汤组神经元受损情况得到改善,神经元的形态结构优于模型组;免疫荧光检测结构显示与假手术组相比,脊髓损伤后GFAP表达显著,损伤处GFAP阳性细胞数增多,差异有统计学意义(P<0.05);与模型组相比,活血通督汤组GFAP阳性细胞数较少,差异有统计学意义(P<0.05)。结论:活血通督汤能促进脊髓损伤后肢体运动功能的恢复,其机制可能与抑制GFAP表达、降低脊髓损伤后胶质瘢痕形成有关。
Objective: To observe the effect of Huoxuetongtang on locomotor function and glial scar formation after spinal cord injury, and to explore the mechanism of action of Huoxuetongtang on spinal cord injury. Methods: Eighteen adult Sprague-Dawley rats were randomly divided into sham-operation group, model group and Huoxutongsuotang group by random number table method, with 6 rats in each group. The sham operation group was treated by laminectomy only. The model group and Huoxue Tongdu Decoction group were given spinal cord injury model with NYU spinal cord striker. After the model was established, the rats in the sham operation group and model group were treated with saline, Tong Du Tang gavage. BBB limb motor function score on the 1st, 3rd, 5th and 7th days after operation, and the material was taken after 7 days. The morphology and structure of neurons were observed by Nissl staining. The expression of glial fibrillary acidic protein (GFAP) was detected by immunofluorescence. The number of GFAP positive cells was calculated. Results: From the third day after operation, the BBB score of rats in Huoxuetongtu group was higher than that of BBB rats in model group (P <0.05). The results of Nissl staining showed that Huoxuetongtang group Compared with the sham-operation group, the expression of GFAP in the injured spinal cord was significantly increased and the number of GFAP-positive cells in the injury increased compared with the sham operation group, the difference was statistically significant (P <0.05). Compared with the model group, the number of GFAP positive cells in Huoxue Tongdu Decoction group was less, the difference was statistically significant (P <0.05). Conclusion: Huoxue Tong Du Tang can promote the recovery of limb motor function after spinal cord injury. The mechanism may be related to the inhibition of GFAP expression and the decrease of glial scar formation after spinal cord injury.
