建立了纸上荧光成像-数码比色法快速测定维生素B2(VB2)的新方法。将VB2溶液点样在滤纸上,在紫外灯下拍摄VB2样点的荧光图像,成像斑点的灰度积分值与VB2的浓度在一定范围内具有线性关系。对点样体积、不同面积色块截取、同一面积色块不同取点位置对灰度积分值的影响进行了考察和优化,在优化的条件下,方法在10~60μg/mL范围内具有良好的线性关系(R2=0.996),对VB2的最低响应值为1.0μg/mL。方法应用于实际样品分析的结果与荧光光谱法一致。标准加入法的回收率为94.1%~102.1%,相对标准偏差在0.5%~2.9%范围。方法可进行现场快速定量测定。
A new method of rapid determination of vitamin B2 (VB2) by fluorescence imaging on paper was developed. The VB2 solution was spotted on the filter paper and the fluorescence image of VB2 sample was taken under ultraviolet light. The gray integral value of imaging spots and VB2 concentration had a linear relationship within a certain range. The effect of spot size, different area patches, different patch locations of the same area on the integral value of the gradation was investigated and optimized. Under the optimal conditions, the method has good performance in the range of 10 ~ 60μg / mL The linear relationship (R2 = 0.996) and the lowest response to VB2 was 1.0 μg / mL. The results of the method applied to the actual sample analysis are consistent with the fluorescence spectroscopy.
