研究了5种标准蛋白在流动相中含有不同脲浓度条件下的疏水色谱保留行为。当脲 浓度不变时,蛋白质的保留仍然服从计量置换保留模型,并可测定在该特定脲浓度条件下蛋白质的Z值。计量置换参数Z可作为疏水色谱中生物大分子构象变化的表征。蛋白质的Z值随脲浓度的增大而减小。除分子构象变化的因素外,Z值变化与脲参与置换剂分子间的计量置换过程有关。Z值变化的连续性与否直接反映了蛋白质分子构象的连续性变化或突变。
Hydrophobic chromatographic retention behavior of five standard proteins was studied in the presence of different concentrations of urea in the mobile phase. When the urea concentration is constant, the protein retention still follows the metered replacement retention model and the Z value of the protein at that particular urea concentration can be determined. The parameter Z can be used to characterize the conformational changes of biological macromolecules in hydrophobic chromatography. The Z value of protein decreased with the increase of urea concentration. In addition to the molecular conformational changes in the factors, the Z value changes and urea involved in the replacement of molecules between the metered displacement process. The continuity of the Z value changes directly reflects the conformational continuity or mutation of the protein molecule.
