目的:研究益肾骨康方对骨转移癌疼痛大鼠脊髓背角胶质原纤维酸性蛋白(GFAP),c-fos,P物质蛋白表达的调控作用。方法:建立裸鼠肺腺癌溶骨性骨转移模型,采用免疫组织化学染色法分别观察各组裸鼠脊髓背角GFAP,c-fos,P物质等蛋白的表达,使用MIA4.0分析软件进行图像分析,每张切片取5个视野,显微镜放大倍数为400倍,统计每个高倍镜视野中表达的阳性物质的光密度值。结果:模型组裸鼠发生了100%的骨转移,免疫组织化学结果显示模型组脊髓背角GFAP,c-fos,P物质蛋白过表达;而益肾骨康方低剂量组与中西药组裸鼠未发生骨转移,免疫组织化学结果显示GFAP,c-fos,P物质蛋白的表达处于较低水平;益肾骨康方中、高剂量组裸鼠也发生了骨转移,但这两组裸鼠脊髓背角GFAP,c-fos,P物质蛋白等与疼痛相关的蛋白表达明显低于模型组。结论:益肾骨康方可通过抑制裸鼠脊髓背角GFAP,c-fos,P物质蛋白的表达而起到一定镇痛作用。
OBJECTIVE: To study the regulatory effect of YKGG on protein expression of glial fibrillary acidic protein (GFAP) and c-fos, P in spinal dorsal horn in a rat model of bone metastatic pain. Methods: To establish osteolytic bone metastasis model of lung adenocarcinoma in nude mice. Expressions of GFAP, c-fos and substance P in spinal dorsal horn of nude mice were observed by immunohistochemical staining. MIA4.0 software Image analysis, taking 5 fields per slice, the magnification of the microscope is 400 times, and the optical density values of the positive substances expressed in each high magnification field are counted. Results: 100% bone metastasis was observed in nude mice in model group. The expression of GFAP, c-fos and substance P in spinal cord dorsal horn in model group was over-expressed by immunohistochemistry. Bone metastasis was not found in the mice. The expression of GFAP, c-fos, and P protein was lower at the immunohistochemical staining. Bone metastasis was also found in the middle and high dose Yishen Gukang decoction groups. However, Pain-related protein expression of GFAP, c-fos and substance P in spinal cord dorsal horn in rats was significantly lower than that in model group. Conclusion: YISHUKANGKANG decoction can inhibit the expression of GFAP, c-fos and P protein in spinal dorsal horn of nude mice.
