目的:探讨胃癌和癌旁组织中Artemin和MMP-9的表达情况及其与浸润转移的关系,分析两种蛋白表达之间的关系。方法:应用免疫组化EnVision法检测80例胃癌组织和20例癌旁组织(距肿瘤边缘>5cm)中Artemin和MMP-9的表达,并分析其与胃癌临床病理特征的关系。结果:胃癌组织Artemin和MMP-9蛋白阳性表达率分别为78.75%(63/80)和68.75%(55/80),明显高于癌旁组织的10.00%(2/20)和15.00%(3/20),差异有统计学意义(χ2=33.242,P=0.000;χ2=18.976,P=0.000)。胃癌组织Artemin和MMP-9蛋白阳性表达均与临床分期、浸润深度及淋巴结转移有关(P<0.05),而与患者年龄、性别、肿瘤大小及分化程度无关,P>0.05。两者阳性表达呈正相关,r=0.705,P=0.000。结论:Artemin和MMP-9蛋白高表达均与胃癌的发生及浸润转移密切相关,联合检测Artemin和MMP-9蛋白表达情况可能成为评估胃癌患者预后指标及潜在肿瘤生物学治疗的新靶点。
OBJECTIVE: To investigate the expression of Artemin and MMP-9 in gastric cancer and paracancerous tissues and its relationship with invasion and metastasis, and analyze the relationship between the expression of two proteins. Methods: The expression of Artemin and MMP-9 in 80 cases of gastric cancer and 20 cases of paracancerous tissues (> 5 cm away from the edge of the tumor) were detected by EnVision immunohistochemistry. The relationship between them was analyzed with clinicopathological features. Results: The positive rates of Artemin and MMP-9 in gastric cancer tissues were 78.75% (63/80) and 68.75% (55/80), respectively, which were significantly higher than those in paracancerous tissues (10.00%, 2/20 and 15.00%, 3 / 20), the difference was statistically significant (χ2 = 33.242, P = 0.000; χ2 = 18.976, P = 0.000). The positive expressions of Artemin and MMP-9 in gastric cancer tissues were correlated with clinical stage, depth of invasion and lymph node metastasis (P <0.05), but not with age, sex, tumor size and differentiation degree (P> 0.05). The positive expression of the two was positively correlated, r = 0.705, P = 0.000. Conclusion: The high expressions of Artemin and MMP-9 are closely related to the carcinogenesis and invasion and metastasis of gastric carcinoma. Combined detection of Artemin and MMP-9 protein expression may be a new target for evaluating the prognosis of gastric cancer patients and potential tumor biological therapy.
