目的探讨原癌基因miR-21在不同年龄段人群血清中表达量的变化及与寿命的关系。方法选取2014年1月至2016年12月间首都医科大学附属北京友谊医院体检及住院的29例不同年龄段人群为研究对象,其中青年组10例,中老年组9例,长寿组10例,提取3组人员的血清样品,利用实时荧光定量聚合酶链反应(PCR)法检测原癌基因miR-21表达水平,判断miR-21在不同年龄段人群血清中的表达量情况。结果长寿组人员血清中miR-21相对表达量为(0.82±0.39),显著低于中老年组的(1.43±0.51),差异有统计学意义(P<0.05)。将肌酐清除率、EF值和血脂水平分别与miR-21相对含量进行比对发现,不同年龄段miR-21相对含量趋势与血脂水平近似,长寿组人群中血脂水平较中老年组人群略有下降,但差异无统计学意义(P>0.05)。结论原癌基因miR-21可能在长寿人群中表达量减低,抑制恶性肿瘤发生,其表达量与血脂有相关性,有望成为寿命相关性基因之一。
Objective To investigate the relationship between the expression of miR-21 in serum of different age groups and its longevity. Methods From January 2014 to December 2016, 29 people of different age groups in Beijing Friendship Hospital affiliated to Capital Medical University were enrolled in this study. Among them, 10 were young group, 9 were middle-aged and elderly group, 10 were longevity group, Serum samples of three groups were extracted and the expression of miR-21 was detected by real-time fluorescence quantitative polymerase chain reaction (PCR) to determine the expression level of miR-21 in serum of different age groups. Results The relative expression of miR-21 in serum of longevity group was (0.82 ± 0.39), which was significantly lower than that of middle aged group (1.43 ± 0.51), the difference was statistically significant (P <0.05). Comparing creatinine clearance rate, EF value and lipid level with the relative content of miR-21 respectively, we found that the trend of relative content of miR-21 in different age groups is similar to that of serum lipids, and the serum lipids in longevity group is slightly lower than that in middle- , But the difference was not statistically significant (P> 0.05). Conclusions The expression of oncogene miR-21 may be decreased in longevity population and inhibited the occurrence of malignant tumors. The expression level of miR-21 may be related to the level of lipids, which is expected to become one of the life-related genes.
