目的探讨妊娠特异性糖蛋白对人外周血树突状细胞(DC)的免疫调节作用。方法常规分离、培养外周血树突状细胞,一定浓度妊娠特异性糖蛋白作用后,采用三色抗体(Lin1-FITC、抗HLA-DR-Percp和PE标记的CD80、CD86、CD11c和CD123抗体)流式检测DC的表型;流式细胞仪检测DC摄取抗原PE-OVA的能力;MTT法检测DC刺激同种T淋巴细胞的增殖能力。结果与对照组比较,妊娠特异性糖蛋白作用后的DC膜表面分子CD80、CD86和CD11c的阳性百分率明显降低(P<0.01),而CD123的阳性率明显升高(P<0.01);妊娠特异性糖蛋白作用后的DC的抗原摄取能力及刺激同种T细胞的增殖能力均明显降低(P<0.01)。结论妊娠特异性糖蛋白对外周血树突状细胞具有免疫下调作用。
Objective To investigate the immunomodulatory effect of pregnancy-specific glycoprotein on human peripheral blood dendritic cells (DCs). Methods After routine isolation and culture of peripheral blood dendritic cells and certain concentrations of pregnancy-specific glycoprotein, three-color antibody (Lin1-FITC, anti-HLA-DR-Percp and PE labeled CD80, CD86, CD11c and CD123 antibodies) Flow cytometry was used to detect the phenotype of DC. Flow cytometry was used to detect the ability of DC to ingest antigen PE-OVA. MTT assay was used to detect the proliferation of allogeneic T lymphocytes stimulated by DC. Results Compared with the control group, the positive percentage of CD80, CD86 and CD11c on the surface of DCs after pregnancy-specific glycoprotein was significantly decreased (P <0.01), while the positive rate of CD123 was significantly increased (P <0.01) The antigen uptake capacity of DCs stimulated by glycoprotein and the proliferation of allogeneic T cells were significantly decreased (P <0.01). Conclusion Pregnancy-specific glycoprotein has immunosuppressive effect on peripheral blood dendritic cells.
