体外实验证实,熊果酸可抑制人类多种肿瘤细胞的增殖并可诱导肿瘤细胞的凋亡,而对活体动物的抗肿瘤作用鲜有报道。本研究利用二甲基肼诱导大鼠大肠癌模型,同时给予含不同剂量的熊果酸饲料喂养,观察并检测肿瘤的形成部位、数量、体积,并用免疫组织化学方法检测大鼠大肠癌组织中COX-2、Bcl-2和Bax表达的变化,对这些指标进行定性及半定量分析。结果显示,熊果酸可抑制肿瘤的形成,呈剂量依赖性抑制癌组织COX-2的表达,但对Bcl-2、Bax的表达无显著影响。熊果酸对大肠癌有防治作用,其机制可能是通过降低COX-2的表达而实现的。
In vitro experiments confirmed that ursolic acid can inhibit the proliferation of a variety of human tumor cells and induce apoptosis of tumor cells, and the antitumor effects on living animals have rarely been reported. In this study, dimethyl hydrazine induced rat model of colorectal cancer, while giving different doses of ursolic acid feed to feed, observe and detect the formation of tumor sites, quantity and volume, and immunohistochemical detection of rat colorectal cancer tissue COX-2, Bcl-2 and Bax expression changes in these indicators of qualitative and semi-quantitative analysis. The results showed that ursolic acid inhibited tumor formation and inhibited COX-2 expression in a dose-dependent manner, but had no significant effect on the expression of Bcl-2 and Bax. Ursolic acid has a preventive and therapeutic effect on colorectal cancer, the mechanism may be achieved by reducing the expression of COX-2.
