整合蛋白α 5β 1是一类存在于细胞表面的细胞粘附分子,在肝癌的发生发展和演进中起着重要的作用。研究了快速定量检测整合蛋白α 5β 1 mRNA表达的 RT- PCR方法,结果表明:在 PCR反应体系中加入 25 ng到 200 ng mRNA的逆转录产物呈较好的线性关系,并以此为反应条件测定 pcDNA3-α 5, pcDNA3-β 1质粒共转染或单转染的α 5β 1.6- 7721, α 5.3- 7721, β 1.6- 7721细胞株 mRNA的表达,表明经 pcDNA3-α 5, pcDNA3-α 1质粒感染后人肝癌细胞株 SMMC7721细胞中整合蛋白α 5β 1的表达有较大幅度的提高,而且α5,β1亚基间可能存在相互的诱导作用。
Integrin α 5β 1 is a type of cell adhesion molecule present on the cell surface and plays an important role in the development and progression of hepatocellular carcinoma. The RT-PCR method for rapid and quantitative detection of integrin α 5β 1 mRNA expression was studied.
