目的考察经口给予纳米骨架给药系统中载体的荷电性对荷电药物缬沙坦体外释放的影响,以期寻找到经口给予纳米骨架给药系统中荷电药物的释放规律。方法采用溶剂挥发法制备不同处方的口服纳米骨架给药系统,用桨板法测定药物释放度,应用X射线衍射法、扫描电镜法、透射电镜法和红外吸收光谱法对其进行表征。结果调节载体材料种类和比例可以控制药物释放速度和程度。当药物与载体带有相反电荷时,加入电解质使药物释放增加;当药物与载体带有相同电荷时,加入电解质使药物释放减少。结论药物与载体之间的电荷作用对经口给予纳米骨架系统中药物的体外释放行为具有重要的影响,可为制备荷电药物的经口给予纳米骨架系统提供参考。
OBJECTIVE To investigate the influence of the electronegativity of carriers on the release of charged drug Valsartan in orally administered nanoshelf drug delivery system in order to find out the regularity of the release of charged drugs in the drug delivery system. Methods Oral nanoshelf drug delivery systems with different prescriptions were prepared by solvent evaporation method. Drug release was determined by paddle method. The drug release was characterized by X-ray diffraction, scanning electron microscopy, transmission electron microscopy and infrared absorption spectroscopy. Results Adjusting the type and ratio of carrier materials can control the rate and extent of drug release. When the drug and the carrier with the opposite charge, the electrolyte added to the drug release increased; when the drug and the carrier with the same charge, adding electrolyte to reduce drug release. Conclusion The charge interaction between drug and carrier has an important effect on the in vitro release behavior of drugs in the nanostructured system. It can provide a reference for the preparation of drug-loaded nanostructures by oral administration.
