目的观察葡萄籽提取物原花青素(GSPE)对三氧化二砷(As2O3)所致肝脏毒性的预防保护效果,揭示其可能机制。方法将84只昆明种小鼠,随机分为35 d空白组、70 d空白组、35 d GSPE组、70 d GSPE组、砷染毒组、GSPE预防组和GSPE干预组,每组12只,雌雄各半,以GSPE 400 mg/kg、As2O35 mg/kg为剂量灌胃给药。测定小鼠体重、肝重、肝脏系数、肝匀浆中丙二醛(MDA)含量、谷胱甘肽(GSH)含量、超氧化物歧化酶(SOD)活力、总抗氧化能力(T-AOC)水平,血清中天冬氨酸转氨酶(AST)与丙氨酸转氨酶(ALT)活力。结果砷染毒组ALT和AST活性、MDA含量显著高于70 d空白组,SOD活性、GSH含量、T-AOC水平显著低于70 d空白组(P<0.05);砷染毒组与GSPE干预组、70 d GSPE预防组相比,血清ALT、AST活力增高(P<0.05);70 d GSPE预防组ALT及AST活性、MDA含量均低于35 d GSPE干预组(P<0.05),SOD活性、GSH含量、T-AOC水平均高于35 d GSPE干预组(P<0.05);GSPE干预组和GSPE预防组较砷染毒组,SOD活力、T-AOC水平、GSH含量均升高,MDA含量均降低(P<0.05)。结论砷染毒前35 d对小鼠进行GSPE预防性干预,可提高肝脏组织抗氧化能力,有效减轻砷致肝脏毒性作用。
Objective To observe the preventive and protective effects of grape seed extract proanthocyanidins (GSPE) on hepatic toxicity induced by As 2 O 3 and to reveal its possible mechanism. Methods A total of 84 Kunming mice were randomly divided into three groups: 35 days blank group, 70 days blank group, 35 days GSPE group, 70 days GSPE group, arsenic exposure group, GSPE prevention group and GSPE intervention group. The activities of ALT, AST and MDA in the GSPE prevention group on the 70 d were lower than those on the GSPE intervention group for 35 d (P <0.05), while the activity of SOD , GSH content and T-AOC level were higher than those of GSPE intervention group (P <0.05). GSPE intervention group and GSPE prevention group were higher than that of arsenic exposure group, SOD activity, T-AOC level, Content decreased (P <0.05). Conclusion Pretreatment with GSPE for mice 35 days before arsenic exposure can improve the anti-oxidative ability of liver tissue and ameliorate the hepatotoxic effects of arsenite.
