为探讨p53和c-myc基因异常表达与膀胱癌多药耐药性(MDR)的关系,应用ABC免疫组织化学方法检测67例膀胱癌癌细胞中p53和c-myc的表达产物及多药耐药基因(mdr-1)产物P-gp。结果显示p53突变蛋白蓄积的膀胱癌中P-gp阳性表达率92.3%;p53阴性者P-gp阳性率为36.6%。P-gp表达与p53突变蛋白蓄积呈显著正相关(r=0.64,P<0.05);而c-myc和mdr-1的表达无明显相关。提示:p53异常表达可增加mdr-1基因的表达,使膀胱癌细胞获得MDR表型
To investigate the relationship between abnormal expression of p53 and c-myc genes and multidrug resistance (MDR) in bladder cancer, the expression products and multidrug resistance of p53 and c-myc in 67 bladder cancer cells were detected by ABC immunohistochemistry. The results showed that the positive expression rate of P-gp was 92.3% in the bladder cancer with accumulation of p53 mutant protein; the positive rate of P-gp in p53-negative patients was 36.6%. There was a significant positive correlation between the expression of P-gp and the accumulation of p53 mutein (r=0.64, P<0.05). However, there was no significant correlation between the expression of c-myc and mdr-1. Tip: Abnormal expression of p53 can increase the expression of mdr-1 gene and make bladder cancer cells acquire MDR phenotype.
