目的 :为评估 As2 O3诱导类风湿性关节炎 (RA)滑膜细胞凋亡在 RA治疗中的作用。方法 :用光镜、电镜、流式细胞仪及 DNA凝胶电泳等方法检测了 As2 O3 诱导体外培养 9例 RA及对照组的 5例骨性关节炎 (OA)与 4例正常人 (NC)关节滑膜细胞凋亡情况。结果 :加药前 RA、OA和NC组凋亡细胞百分率分别是 3.8%± 0 .7% ,0 .8%± 0 .3%和 0 ,加药后均明显升高 ,RA:61 .1 %± 9.7% (P<0 .0 1 ) ,OA:33.3%± 4.6% (P<0 .0 1 ) ,NC:1 7.3%± 1 .1 % (P<0 .0 1 )。RA组显著高于 OA组 (P<0 .0 1 )和 NC组 (P<0 .0 1 )。结论 :As2 O3可诱导 RA滑膜细胞凋亡并且可能因此而起治疗作用 ,同时可为临床筛选 RA治疗用药及剂量提供帮助
AIM: To evaluate the role of As2 O3 in the induction of rheumatoid arthritis (RA) synoviocyte apoptosis in RA. Methods: Five cases of osteoarthritis (OA) and four normal controls (NC) were treated with As2O3 in 9 RA and control groups by light microscope, electron microscope, flow cytometry and DNA gel electrophoresis. Synovial cell apoptosis. Results: The percentages of apoptotic cells in RA, OA and NC groups before treatment were 3.8% ± 0.7%, 0.8% ± 0.3% and 0% % ± 9.7% (P <0.01), OA: 33.3% ± 4.6% (P <0.01), NC: 1 7.3% ± 1.1% (P <0.01). RA group was significantly higher than OA group (P <0.01) and NC group (P <0.01). Conclusion: As2 O3 can induce the apoptosis of RA synovial cells and may play a therapeutic role in this process. At the same time, it can be helpful for the clinical screening of RA therapeutic dosage and dose
