以CAS-200型图象细胞仪(ICM)对99例肝细胞癌(HCC)行DNA定量分析。结果:包膜完整、包膜不完整、和无包膜三组HCC之间,增殖指数、细胞核面积、二倍体肿瘤比例均有显著的统计学差异(P<0.01,P<0.05,P<0.05)。在无门静脉癌栓(TTPV)、合并镜下TTPV、和合并肉眼TTPV的三组HCC间,增殖指数和DNA指数统计学差异显著(P<0.01,P<0.05)。合并肝内播散组HCC,增殖指数及超五倍体细胞比例分别显著高于无肝内播散组(P<0.05,P<0.01)。然而组间比较表明,包膜不完整组与无包膜组之间,合并镜下TTPV组与合并肉眼TTPV组之间,ICM的各项参数均无统计学差异。结论:①随着DNA增殖水平的上升,HCC细胞获得了更强的侵袭性和转移能力;②以ICM行DNA定量分析对于揭示HCC是否具有更强侵袭性和转移潜能有明显价值,其中增殖指数检测具有效高的敏感性;③进一步研究ICM的各项参数对HCC预后的价值是有意义的。
A total of 99 hepatocellular carcinoma (HCC) lines were quantitatively analyzed by a CAS-200 image cytometer (ICM). RESULTS: There were significant differences in the proliferative index, nuclear area, and diploid tumor ratio between the intact capsules, incomplete capsules, and non-enveloped HCCs (P<0.01, P<0. 05, P<0.05). In the three groups of HCC without portal vein tumor thrombosis (TTPV), combined TTPV, and macroscopic TTPV, the difference in proliferation index and DNA index was statistically significant (P<0.01, P<0.05). With the intrahepatic dissemination group HCC, the proliferation index and the proportion of ultra-quintile cells were significantly higher than those without intrahepatic dissemination group (P<0.05, P<0.01). However, comparisons between groups showed that there were no statistically significant differences in ICM parameters between the incompletely encapsulated group and the non-enveloped group and between the combined TTPV group and the macroscopic TTPV group. Conclusions:1 With the increase of DNA proliferation, HCC cells acquire more invasiveness and metastasis capacity;2 The quantitative analysis of DNA by ICM has significant value in revealing whether HCC has stronger invasiveness and metastatic potential, in which the proliferation index Detection has a high sensitivity; 3 further study of the parameters of ICM on the value of HCC is meaningful.
