目的:建立LL/2-Luc-M38小鼠肺癌骨转移模型,探讨补肾散结方对肺癌骨转移及PTHr P、TGF-β1表达的影响。方法:在C57小鼠骨髓腔接种LL/2-Luc-M38肺癌细胞株建立骨转移模型,并随机分为模型组、帕米膦酸钠组、补肾散结组。模型组以生理盐水灌胃,帕米膦酸钠组腹腔注射帕米膦酸钠(0.1 ml/次),补肾散结组小鼠以补肾散结方药液灌胃。观察小鼠X线股骨表现和骨转移细胞IVIS荧光信号,骨肿瘤组织病理,骨组织PTHr P、TGF-β1蛋白表达和mRNA表达情况。结果:模型组见明显溶骨性骨破损、肿瘤细胞密集,补肾散结组和帕米膦酸钠组骨皮质轻度破坏、肿瘤细胞减少;帕米膦酸钠组和补肾散结组骨组织肿瘤细胞荧光信号表达量均低于模型组(P<0.05,P<0.01);帕米膦酸钠组、补肾散结组骨组织PTHr P、TGF-β1mRNA表达均明显低于模型组(P<0.05,P<0.01)。结论:补肾散结方可能通过抑制TGF-β1、PTHr P蛋白和mRNA表达达到抗肿瘤骨转移的作用。
OBJECTIVE: To establish a model of lung cancer bone metastasis in LL / 2-Luc-M38 mice and explore the effect of Bushen Sanjie Recipe on the bone metastasis of lung cancer and the expression of PTHr P and TGF-β1. Methods: Bone metastasis was induced in LL / 2-Luc-M38 lung cancer cell line in C57 mouse bone marrow cavity and randomly divided into model group, pamidronate group and Bushen Sanjie group. The model group was given gavage with saline, and the pamidronate sodium group was injected intraperitoneally with sodium pamidronate (0.1 ml / time). The X-ray appearance of femur, the IVIS fluorescence signal of bone metastasis cells, the histopathology of bone tumor, the expression of PTHr P, the expression of TGF-β1 protein and the mRNA expression in bone were observed. Results: In the model group, the osteolytic bone was obviously damaged, the tumor cells were dense, the cortical bone was destroyed slightly in Bushen Sanjie group and Pamidronate group, and the number of tumor cells was decreased. In Pamidronate group and Bushen Sanjie group, (P <0.05, P <0.01). The expression of PTHr P and TGF-β1mRNA in pamidronate and Bushen Sanjie group were significantly lower than those in model group (P < 0.05, P <0.01). Conclusion: Bushen Sanjie decoction may inhibit the bone metastasis of tumor by inhibiting the expression of TGF-β1 and PTHr P protein and mRNA.
