采用细胞融合和单克隆技术,获得抗羟甲芬太尼(ohmefentanyl,Ohm)单克隆抗体MAb-D_2和MAb-F_1·MAb-D_2和MAb-F_1与Ohm-BSA结合亲和力高、专一性强。MAh-D_2和MAb-F_1还能抑制[~3H]Ohm与大鼠脑匀浆膜蛋白上的阿片受体的结合,并能对抗Ohm抑制电场刺激引起豚鼠回肠收缩的作用。以上结果表明MAb-D_2和MAb-F_1是特异性的抗Ohm单克隆抗体,作为Ohm的拮抗剂具有广泛用途。
By using cell fusion and monoclonal technology, the monoclonal antibodies MAb-D_2, MAb-F_1 · MAb-D_2 and MAb-F_1 of Ohme monoclonal antibody were obtained. The binding affinity of Ohm-BSA was high, . MAh-D_2 and MAb-F_1 also inhibited the binding of [~ 3H] Ohm to opioid receptors in the membrane proteins of rat brain homogenates and were able to counteract the contractile effect of Ohm on the ileum of guinea pigs induced by electric field stimulation. The above results indicate that MAb-D 2 and MAb-F 1 are specific anti-Ohm monoclonal antibodies and have wide range of uses as antagonists of Ohm.
