目的观察DON毒素与氟致骨软骨损伤的联合作用机制中,对转化生长因子β2(TGF-β2)、成纤维细胞生长因子23(FGF-23)等4种骨生长发育相关的指标的影响,探讨DON毒素与氟的联合作用和茶的拮抗效应。方法 28日龄Wistar大鼠幼鼠30只,按照完全随机化方法分空白组、“DON+F”组和“DON+F+茶”组。空白组饮用超纯水,“DON+F”组饮用150mg/L NaF超纯水溶液;“DON+茶+F”组饮用由茶水溶解的150mg/L NaF水溶液;DON毒素染毒组各组按0.2μg/g DON毒素灌胃,空白组用等量超纯水灌胃,每天1次。喂养5周后,麻醉后断头取血,ELISA法测定各细胞因子。结果 ELISA结果显示,血清中TGF-β2的平均含量“DON+F”组(55.47pg/ml)显著高于对照组(29.2 pg/ml)和“DON+F+茶”组(38.4 pg/ml);FGF-23的平均含量“DON+F”组(205.49pg/ml)显著高于对照组(192.92 pg/ml)和“DON+F+茶”组(97.3pg/ml);BMP-2的平均含量“DON+F”组(15.24ng/ml)显著高于对照组(12.44ng/ml)和“DON+F+茶”组(12.9ng/ml);TNF-α的平均含量“DON+F”组(936.88pg/ml)显著高于对照组(579.54 pg/ml)和“DON+F+茶”组(652.95 pg/ml)。结果提示DON与F可导致骨软骨损伤,茶具有保护作用。结论茶可在一定程度上可减轻DON毒素和氟对骨软骨的损伤,显示了一定的拮抗作用,但不能消除DON毒素和F的致病效应。
