目的:从肌肉蛋白质组表达探讨“芪枣四君汤”提高大强度训练机体运动耐力的机制。方法:将ICR小鼠50只分为安静对照组(C)、运动训练组(T)和中药运动组(Z)。T和Z组进行逐级递增负荷跑台训练4周后力竭运动和双向凝胶电泳(2-DE)分离腓肠肌蛋白,清华紫光D3000扫描凝胶和PD quest软件分析。结果:游泳至力竭的时间Z组显著长于T组(P<0.05)。T组与C组间有31个蛋白质点变化,有2个上调2倍以上,5个下调2倍以上;Z组与T组有28个蛋白质点变化,有4个上调2倍以上,有3个下调2倍以上。蛋白质点5703和4301表达C组高、T组低和Z组更低,点7201和1201的表达C与T组高、Z组低。结论:训练小鼠在“芪枣四君汤”干预下运动耐力提高和骨骼肌蛋白质组发生差异表达,差异蛋白可能参与了工作能力提高过程。
OBJECTIVE: To explore the mechanism of improving the exercise endurance of the body by intensive training from the muscle proteome expression. Methods: 50 ICR mice were divided into quiet control group (C), exercise training group (T) and Chinese medicine exercise group (Z). T and Z groups were subjected to progressive treadmill training 4 weeks after exhaustive exercise and two-dimensional gel electrophoresis (2-DE) to separate gastrocnemius proteins, and serum H3000 scanning gel and PD quest software. Results: The swimming time to exhaustion in Z group was significantly longer than that in T group (P <0.05). There were 31 protein spots in T group and C group, with 2 up-regulated more than 2 times and 5 down-regulated more than 2 times. There were 28 protein spots in Z group and T group, 4 up-regulated more than 2 times and 3 A reduction of more than 2 times. Protein spots 5703 and 4301 expressed high in group C, low in group T and lower in group Z. The expression of point 7201 and 1201 was higher in group C than in group T and lower in group Z. CONCLUSION: The training mice showed increased exercise endurance and skeletal muscle proteome expression under the intervention of “Qizao Sijun Decoction”, and the differential proteins may be involved in the improvement of working ability.
