本文报道神经节苷脂单组分GM_2、GM_3、GD_3及脑苷脂对猪肾Na+、K~+-ATP酶活性的影响。实验结果表明,GM_3对Na~+、K~+-ATP酶有明显抑制作用。结构上比GM_3多一个唾液酸的GD_3和多一个乙酰糖基的GM_2均无抑制怍用。即使去除唾液酸的CDH,或是在此基础上再去除一个半乳糖基的脑苷脂(CMH)及唾液酸,对该酶也无抑制作用。提示GM_3结构完整的重要性。动力学研究表明,在不同Na~+、K+浓度对,GM_3对Na~+、K~+-ATP酶的抑制作用,均呈非竞争性抑制。
This article reports the effects of GM2, GM3, GD3 and cerebrosides on the activity of Na +, K + -ATPase in porcine kidney. The experimental results show that GM_3 has obvious inhibitory effect on Na ~ + and K ~ + -ATPase. GD_3, which is one more sialic acid in structure than GM_3, and GM_2, which is more than one acetylglycoside, showed no inhibitory effect. Even removal of sialic acid CDH, or on the basis of the removal of a galactosyl cerebroside (CMH) and sialic acid, the enzyme has no inhibitory effect. Prompts the completeness of the structure of GM_3. Kinetic studies show that, in different Na ~ +, K + concentration, GM_3 Na ~ +, K ~ + -ATP enzyme inhibition were non-competitive inhibition.
