目的观察清肝化痰活血方对非酒精性脂肪性肝炎模型大鼠水通道蛋白7、9(AQP-7、9)表达的影响。方法采用高脂饲料喂养复制模型大鼠,第8周分别处死正常组大鼠5只和模型组大鼠10只,模型组剩余大鼠再分为高脂饲料组、易善复组和清肝化痰活血方组,给药5周后,处死大鼠,留取血清、肝组织标本,检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、三酰甘油(TG)、胆固醇(TC)及肝组织中AQP-7、9的表达;另肝组织经HE染色光镜下观察肝组织病理学变化。结果第8周时,模型组大鼠脂肪变性程度和炎症活动度计分与正常组比较有显著性差异(P<0.01)。第13周时,高脂饲料组大鼠肝细胞脂肪变性和炎症活动度计分与正常组、西药组和中药组相比有显著性差异(P<0.05,P<0.01),肝组织中AQP-7、AQP-9 mRNA的含量与正常组、西药组和中药组相比差异有统计学意义(P<0.01);清肝化痰活血方组大鼠肝组织脂肪变性程度和炎症活动度计分与高脂饲料组比较差异有统计学意义(P<0.05),血清AST、ALT、TG、TC的含量与高脂饲料组比较明显下降(P<0.01),肝组织中AQP-7含量较高脂饲料组明显上升(P<0.01),AQP-9含量较高脂饲料组明显下降(P<0.01)。结论 AQP-7、9与非酒精性脂肪性肝炎的发病有关,清肝化痰活血方可能通过调节AQP-7、9的表达进而改善肝细胞脂质代谢起到防治非酒精性脂肪性肝炎的作用。
Objective To observe the effect of “Qinggan Huatan Huoxue Decoction” on the expression of aquaporin 7,9 (AQP-7,9) in non-alcoholic steatohepatitis model rats. Methods The model rats were fed with high-fat diet. In the 8th week, 5 rats in normal group and 10 rats in model group were sacrificed. The remaining rats in model group were further divided into high fat diet group, After 5 weeks of administration, rats were sacrificed and serum and liver samples were taken for detection of serum ALT, AST, TG, TC, And liver tissue expression of AQP-7,9; the other liver tissue by HE staining under light microscope pathological changes in liver tissue. Results At the 8th week, the scores of steatosis and inflammatory activity in the model group were significantly different from those in the normal group (P <0.01). At week 13, there was a significant difference (P <0.05, P <0.01) in hepatic steatosis and inflammatory activity score in high fat diet group compared with those in normal group, western medicine group and traditional Chinese medicine group, AQP -7, AQP-9 mRNA levels compared with the normal group, western medicine group and traditional Chinese medicine group, the difference was statistically significant (P <0.01); the degree of hepatic steatosis and inflammatory activity meter (P <0.05). The levels of AST, ALT, TG and TC in serum were significantly decreased compared with those in high-fat diet group (P <0.01). The contents of AQP-7 in liver tissue were significantly lower than those in high-fat diet group Fat diet group significantly increased (P <0.01), AQP-9 levels were significantly lower than the high-fat diet group (P <0.01). Conclusions AQP-7,9 is associated with the pathogenesis of non-alcoholic steatohepatitis. Qinggan Huatan Huoxue prescription may improve the lipid metabolism of hepatocytes and prevent non-alcoholic steatohepatitis by regulating the expression of AQP-7,9 effect.
