目的探讨磷脂酸磷酸酶2域1A(PPAPDC1A)在人4种子宫颈癌细胞中表达的差异。方法通过实时荧光定量聚合酶链式反应和Western blot法分别检测PPAPDC1A mRNA和蛋白在人子宫颈癌C33-A细胞、Caski细胞、HeLa细胞和SiHa细胞中的表达。结果子宫颈癌SiHa、HeLa、Caski和C33-A细胞中PPAPDC1A mRNA的表达量分别为0.344±0.027、0.593±0.017、0.899±0.030和2.920±0.045,PPAPDC1A蛋白的表达分别为0.514±0.030、1.185±0.096、1.379±0.084和1.761±0.072;子宫颈癌SiHa、HeLa、Caski和C33-A细胞中PPAPDC1A mRNA和蛋白的表达比较差异有统计学意义(P<0.05)。PPAPDC1A mRNA和蛋白在4种子宫颈癌细胞中表达的顺序为:C33-A细胞>Caski细胞>HeLa细胞>SiHa细胞。结论 PPAPDC1A基因在不同子宫颈癌细胞中存在差异表达。
Objective To investigate the difference of phosphatidic acid phosphatase 2 domain 1A (PPAPDC1A) expression in four human cervical cancer cells. Methods The expression of PPAPDC1A mRNA and protein in human cervical carcinoma C33-A cells, Caski cells, HeLa cells and SiHa cells were detected by real-time fluorescence quantitative polymerase chain reaction and Western blot respectively. Results The expressions of PPAPDC1A mRNA in SiHa, HeLa, Caski and C33-A cells were 0.344 ± 0.027, 0.593 ± 0.017, 0.899 ± 0.030 and 2.920 ± 0.045, respectively. The expression of PPAPDC1A protein in cervical cancer was 0.514 ± 0.030 and 1.185 ± 0.096, 1.379 ± 0.084 and 1.761 ± 0.072, respectively. The expression of PPAPDC1A mRNA and protein in cervical cancer SiHa, HeLa, Caski and C33-A cells was significantly different (P <0.05). The sequence of PPAPDC1A mRNA and protein expression in four kinds of cervical cancer cells was C33-A cells> Caski cells> HeLa cells> SiHa cells. Conclusion The PPAPDC1A gene is differentially expressed in different cervical cancer cells.
