目的:研究紫杉醇对HO8910人卵巢癌细胞的体外杀伤作用;紫杉醇对MMP-2、MMP-9 mRNA表达的影响。方法:采用MTT法检测紫杉醇对HO8910细胞的抑制率;采用半定量RT-PCR法检测紫杉醇作用于HO8910细胞时MMP-2、MMP-9基因mRNA水平的变化。结果:紫杉醇在达10-7mol/L以上浓度时对HO8910细胞抑制率>30%(P<0.01);10-8mol/L以上浓度时紫杉醇可下调MMP-2、MMP-9 mRNA的表达,其作用呈剂量依赖性。结论:在10-7mol/L以上浓度,HO8910细胞对紫杉醇具有药物敏感性;紫杉醇可能通过抑制MMP-2、MMP-9 mRNA的表达,抑制肿瘤的侵袭及转移。
Objective: To study the killing effect of paclitaxel on HO8910 human ovarian cancer cells in vitro and the effect of paclitaxel on the expression of MMP-2 and MMP-9 mRNA. Methods: The inhibitory rate of paclitaxel on HO8910 cells was detected by MTT assay. The mRNA expression of MMP-2 and MMP-9 in paclitaxel-treated HO8910 cells was detected by semi-quantitative RT-PCR. Results: Paclitaxel inhibited the proliferation of HO8910 cells by more than 10-7mol / L (P <0.01). When the concentration of 10-8mol / L or higher, paclitaxel down-regulated the expression of MMP-2 and MMP-9 mRNA The effect was dose-dependent. CONCLUSIONS: HO8910 cells are drug sensitive to paclitaxel at a concentration above 10-7mol / L. Paclitaxel may inhibit tumor invasion and metastasis by inhibiting the expression of MMP-2 and MMP-9 mRNA.
