目的:蛋白激酶C(PKC)抑制剂诱导CNE-2Z细胞凋亡时细胞周期的观察。方法:PKC催化区抑制剂Staurospoine(ST)和调节区抑制剂Sphingosine(SS),终浓度分别为1X10-6mol/L和4X10-5mol/L,诱导CNE-2Z细胞24h,用流式细胞仪(FCM)分析。结果:诱导组细胞周期与对照组比较,ST使细胞G1和S期明显减少及G2期明显增加(P<0.01);SS使S期增加和G2期减少明显(P<0.05),而G1期变化不大(P>0.05)。诱导组G1期前均有明显的亚二倍体峰(凋亡峰)。结论:PKC抑制剂SS、ST诱导CNE-2Z细胞凋亡,同时存在明显的细胞周期异常改变。
AIM: To observe the cell cycle of CNE-2Z cells induced by protein kinase C (PKC) inhibitor. Methods: The CNE-2Z cells were induced by Staurospoine (ST) and Sphingosine (SS), the inhibitor of PKC, at a concentration of 1 × 10-6 mol / L and 4 × 10-5 mol / L, respectively. FCM) analysis. Results: Compared with the control group, the cell cycle in induction group was significantly decreased in G1 and S phase and significantly increased in G2 phase (P <0.01). SS increased the S phase and decreased G2 phase significantly (P <0.05) , But little change in G1 (P> 0.05). The induced group had obvious sub-diploid peak (apoptosis peak) before G1 phase. CONCLUSION: PKC inhibitor SS and ST can induce apoptosis of CNE-2Z cells with obvious changes of cell cycle abnormalities.
